Catalase in Liver


The presence of a specific enzyme can be detected by the reaction it catalyzes, and the enzyme concentration can be measured by the amount of product it forms in a given period of time. Enzyme activity is affected by pH, temperature, and the availability of substrates and coenzymes.

OBJECTIVES:

1. Describe the lock-and-key model of enzyme activity and use this model to explain enzyme specificity.
2. Describe the effect of pH and temperature on enzyme activity.
3. Record observations of enzyme activity to Hydrogen Peroxide.

Catalase is an enzyme that converts hydrogen peroxide to water and oxygen gas. Found in many tissues, catalase is one of the most rapidly acting enzymes in the body (liver).

TO DO LAB YOU NEED THE FOLLOWING MATERIALS:
Small 100-mL beaker
50 mL of hydrogen peroxide (use graduated cylinder to measure the amount specified)
Scissors
Chicken liver
Composition book
Tweezers

PROCEDURE:
1. Using a graduated cylinder, measure 50mL of hydrogen peroxide. For accuracy, locate the meniscus at eye level.
2. Pour 50ml of hydrogen peroxide into a small 100 mL beaker.
3. Mince a fresh chicken liver with scissors.
4. Using the tweezers, immerse chicken liver in the solution (hydrogen peroxide) and gently stir.
5. Record observations in composition book.

Laboratory Report: Put answers into composition book

Describe what occurred when the chicken liver was placed in the beaker of hydrogen peroxide. Write a simple equation that might describe this reaction.
